Stemolecule™ SHH Antagonist is an inhibitor of Sonic hedgehog signaling (SHH)1. Hedgehog signaling plays key roles in a wide variety of development process2 as well as cancer progression3. The vertebrate family of hedgehog genes that have been identified are known as Sonic (Shh), Desert (Dhh), and Indian (Ihh) hedgehogs, all of which encode secreted proteins. Among the three, Sonic hedgehog (SHH) is the best studied ligand of the vertebrate pathway. One of the best studied examples is the action of Sonic hedgehog during development of the vertebrate limb, in which both the concentration and the time of exposure to SHH determines which digit into which the tissue will develop4. Stemolecule™ SHH Antagonist is the result of a compound screening for inhibiting activation of the hedgehog signaling pathway, e.g. inhibition of aberrant growth states resulting from phenotypes such as hedgehog gain-of-function1.

The purity of SHH Antagonist was determined by HPLC analysis. The accurate mass and structure of SHH Antagonist were determined by mass spectrometry and NMR, respectively. Cellular toxicity of SHH Antagonist was tested on mouse ES cells.
