Novel culture medium that is able to maintain high CYP activity in human primary hepatocytes

Summary: Primary hepatocytes rapidly lose their ability to express key metabolic enzymes upon extended culture. This instability causes logistical problems and higher costs for drug screening in vitro assays. In this study, we developed a new cell culture medium that maintains high CYP activities of human primary hepatocytes up to 96 hours in culture. The new formulation shows high performance for in vitro drug metabolism studies with improved CYP expression, induction levels, and stability of expression over time in both 2D and 3D culture formats, while maintaining a highly distinct cobblestone morphology characteristic of mature human hepatocytes.

Functional improvement of iPS cell-derived Hepatocyte (ReproHepatoTM) by utilizing 3D culture system

Summary: Cell immaturity and donor variation are common drawbacks posed by iPS cell-derived hepatocytes. Hence, we have utilized 3D cultivation for iPS cell-derived hepatocytes to address the maturation issue, and multiple iPS cell clones to average out the variation between donors. Maturation of human iPS cell-derived hepatocytes is increased by two different 3D cell culture approaches. Both approaches were evaluated by measuring the expression level of key Cytochrome P450 (CYP) family members. These approaches used in combination may be an attractive model for applications in drug discovery or small molecule evaluations.

iPS-Derived Cells

The world's first iPS cell-derived highly-differentiated human hepatocytes were introduced by ReproCell Inc. These ReproHepato™ cell products are suitable for analysis of a wide variety of hepatic functions, including small molecule drug induction of CYP (Cytochrome P450) isozymes.


ReproCardio 2 cardiomyocytes are delivered as frozen single cells. Using the specially-developed plates included in the kit, you can re-constitute the cells into thin layers or clumps depending on your purpose of research. The re-constituted thin layers and clumps beat periodically and robustly which provide excellent in vitro models of in vivo heart.

Alzheimer Disease cell lines are iPSC-derived neuro progenitor cell lines that are suitable for studying Alzheimer’s disease cell physiology, or use in drug discovery or toxicology assays.  Upon thawing, the cells need to be seeded in ReproNeuro™ Culture medium or ReproNeuro™ MQ medium on a 96-well plate that has been pre-coated with poly-L-Lysine and ReproNeuro™ Coat.  After incubation for approximately 2-weeks, the neuro progenitor cells differentiate into a mixed population of various neuronal cell types including dopaminergic, cholinergic, glutamatergic, and GABAergic neurons.

iPSC-derived neural progenitor cell that came from a 94 year old male patient who had late-stage Type 4 (AD4) familial Alzheimer’s disease.  The frequency of this mutation in early onset Alzheimer’s patients is somewhat rare, at less than 5%.

iPSC-derived neural progenitor cell that originally came from a 20 week old male fetus, but was engineered to resemble Alzheimer’s.  This cell overexpresses a mutant PSEN1 gene giving rise to a genotype and phenotype similar to Type 3 (AD3) familial Alzheimers disease.  The frequency of the PSEN1 genetic mutation in Alzheimer’s disease patients is the major form of EOFAD.
