Characterization is a critical step in confirming status of pluripotent stem cell (iPS cell) cultures during reprogramming and at all stages of differentiation. Since this is a crucial, yet routine process, confirmation of cell status should be simple, quick and above all reliable. The Stemgent™ StainAlive™ antibody format allows for direct staining of live cells in culture and therefore eliminates the need to sacrifice valuable material that can be used for downstream experiments.
Stemgent® StainAlive™ Antibodies
Human H1 ES cells were analyzed live in culture with Stemgent StainAlive DyLight™ 488 TRA-1-60 antibody. Light field microscopy was used to visualize colonies. Direct ICC analysis of live cells in culture allows visualization of pluripotent cells without having to sacrifice valuable wells.
Stemgent Alkaline Phosphatase (AP) Staining Kit II
The state of differentiation can be monitored by detection of AP, along with stem cell markers such as transcription factors Oct-3/4, Nanog, Sox-2 and surface markers SSEA-1, -3, -4, TRA-1-60 and TRA-1-81. Although stem cells from different sources or species display different cell surface markers, AP is a universal marker for all types of pluripotent stem cells. The Stemgent® (AP) Staining Kit II uses an easy, histochemical staining protocol with substrate that is stable for use up to 30 minutes allowing for greater throughput.
