Product Overview

Transforming Growth Factor beta 1 (TGF-β1) is a member of the TGF-β superfamily of multifunctional polypeptide growth factors that includes bone morphogenetic proteins (BMPs), Activin, and Nodal. TGF-β1 both helps maintain pluripotency in human embryonic stem (hES) cells and induces differentiation1,2. It also induces epithelial-mesenchymal transition and suppresses proliferation and differentiation of several types of lymphocytes, thus playing important roles in human cancers3. Stemfactor TGF-β1 is a 25 kDa homodimeric recombinant protein expressed in human HEK293 cells and composed of two identical 112 amino acid polypeptide chains linked by a single disulfide bond.

Purity

Greater than 95% by SDS-PAGE analysis

Endotoxin Level

Less than 1.0 EU/μg of TGF-β1 as determined by the LAL method.

Biologic Activity

The ED50 is less than 0.2 ng/ml as determined by its ability to inhibit the dose-dependent inhibition of IL-4 induced proliferation of mouse HT-2 cells.

Storage and Stability

Stemfactor TGF-β1 is shipped at room temperature. Lyophilized TGF-β1 is stable for up to 6 months from date of receipt when stored at -20°C to -80°C. Reconstituted TGF-β1, at concentrations greater than or equal to 0.1 mg/ml, is stable for up to 3 months when stored at -20°C and up to 6 months when stored at -80°C.

Reconstitution

Centrifuge briefly and then reconstitute TGF-β1 in 4 mM HCl to yield a stock solution of no less than 0.1 mg/ml. Avoid freeze-thaw cycles as it can result in loss of activity.
