Product Overview

Sonic hedgehog (SHH) is used to study differentiation and expansion of human and mouse embryonic and adult stem cells. Studies suggest SHH is involved in regulating stem cell fates of neural and hematopoetic lineages1. It controls cell division of adult stem cells and has been implicated in development of some cancers2. SHH is instrumental in early embryo patterning and development. It has been implicated as the key inductive signal in patterning of the ventral neural tube, the anterior-posterior limb axis, and the ventral somites3. The mature biologically active form of SHH molecule is produced by autocatalytic cleavage of its precursor protein and corresponds to approximately the N-terminal half of the precursor molecule. Sonic Hedgehog is an approximately 20 kDa protein expressed in E. coli consisting of 175 amino acid residues, including an N-terminal Ile-Ile sequence substituted for the naturally occurring chemically modified Cys residue.

Purity

Greater than 98% by SDS-PAGE analysis

Endotoxin Level

Less than 0.1 ng/µg (1 EU/µg) of SHH as determined by the LAL method.

Biologic Activity

The ED50 is less than 1 μg/ml as determined by its ability to induce alkaline phosphatase production by C3H10T1/2 fibroblasts.

Storage and Stability

Stemfactor SHH is shipped at room temperature. Lyophilized SHH is stable for up to 6 months from date of receipt when stored at -20°C to -80°C. Reconstituted SHH, at concentrations greater than 0.1 mg/ml, is stable for up to 3 months when stored at -20°C and for 1 year when stored at -80°C.

Reconstitution

Centrifuge briefly and then reconstitute in 1X PBS to yield a stock solution of no less than 0.1 mg/ml of SHH. Reconstituted protein should be used immediately or stored in working aliquots at -20°C. For long-term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid freeze-thaw cycles as it can result in loss of activity.
